The director would be replacing "Spider-Man: Homecoming" writers John Francis Daley and Jonathan Goldstein who left the project in 2016.
Muschietti will also be producing the film alongside Barbara Muschietti and Michael Disco.
The star of "The Flash" will continue to be Ezra Miller, who has played him in "Batman v Superman: Dawn of Justice," "Suicide Squad" and "Justice League."
